Markers are very much like a short-term loan that the casino provides players with a substantial bankroll. The process to setup the ability to take out a marker with the casino is very similar to applying for a credit card and being given a credit limit.
Only players that already have an account setup with the casino can ask for markers and when they have to pay it back is dependent upon the agreement that they have with the casino. Usually they will take care of paying off the marker at the casino cage/cashier.
As break-in dealers, you will most-likely not have to deal with markers at least for a little while. As a result, whenever you do encounter a player requesting a marker, the most important thing for you to know is that you must call floor once a player makes a marker request.
DEALERS DO NOT HANDLE MARKERS WITHOUT FLOOR
Floor knows that break-in dealers don't have much experience with markers and will walk you through the process when one comes up. Simply follow floor's instructions and all will be well.
